Antisense Transcripts and Antisense Protein: A New Perspective on Human Immunodeficiency Virus Type 1

چکیده
It was first predicted in 1988 that there may be an Open Reading Frame (ORF) on the negative strand of Human Immunodeficiency Virus type 1 (HIV-1) genome could encode a protein named AntiSense Protein (ASP). In spite some controversy, reports began to emerge years later describing detection HIV-1 antisense transcripts, presence ASP transfected and infected cells, existence immune response targeting ASP. Recently, it established asp gene is exclusively conserved within pandemic group M HIV-1. this review, we summarize latest findings transcripts ASP, discuss their potential functions infection together with role played by ASPs other viruses. Finally, suggest pathways raised study warrant exploration future.

In vitro effect of antisense oligonucleotides on human immunodeficiency virus type 1 reverse transcription.
The molecular events involved in antisense-mediated inhibition of retroviral transcription were studied by analyzing the in vitro effect of antisense oligodeoxynucleotides on reverse transcription by Human Immunodeficiency Virus type 1 (HIV-1) reverse transcriptase (RT). Oligonucleotides have been designed to be complementary to three targets located in the 5' region of the HIV-1 RNA genome: th...

Antisense RNA, transcribed intracellularly from constitutive expression cassettes, inhibits the replication of the human immunodeficiency virus type 1 (HIV-1) as demonstrated by a quantitative microinjection assay in human SW480 cells. Infectious proviral HIV-1 DNA was co-microinjected together with a fivefold molar excess of plasmids expressing antisense RNA complementary to a set of ten diffe...

The antiviral activities of intracellularly expressed antisense RNAs complementary to the human immunodeficiency virus type 1 (HIV-1) pol, vif, and env genes and the 3' long terminal repeat (LTR) sequence were evaluated in this comparative study. Retroviral vectors expressing the antisense RNAs as part of the Moloney murine leukemia virus LTR promoter-directed retroviral transcript were constru...
متن کاملNatural antisense transcripts.
Recent years have seen the increasing understanding of the crucial role of RNA in the functioning of the eukaryotic genome. These discoveries, fueled by the achievements of the FANTOM, and later GENCODE and ENCODE consortia, led to the recognition of the important regulatory roles of natural antisense transcripts (NATs) arising from what was previously thought to be 'junk DNA'. Roughly defined ...
متن کاملResistance mechanism of human immunodeficiency virus type-1 protease to inhibitors: A molecular dynamic approach
Human immunodeficiency virus type 1 (HIV-1) protease inhibitors comprise an important class of drugs used in HIV treatments. However, mutations of protease genes accelerated by low fidelity of reverse transcriptase yield drug resistant mutants of reduced affinities for the inhibitors. This problem is considered to be a serious barrier against HIV treatment for the foreseeable future. In this st...
